Elaboration of a Toxigenic Vibrio cholerae Typing Scheme Based on Bioinformatiсs Analysis Data

Introduction: The current stage of the seventh cholera pandemic is characterized by emergence novel Vibrio cholerae gene variants, gradually replacing their predecessors and occupying a dominant position in etiology disease. Determining epidemic potential identifying number genetic markers unsuitable for operational analysis. Thus, development method differentiating pathogens based on PCR detection limited seems relevant. Objective: To create database whole genome V. sequences containing different alleles cheA3 (VCA1095) rtxA genes bioinformatics analysis data to elaborate simple informative toxigenic vibrio typing scheme. Materials methods: NCBI database-extracted results sequencing 3,309 strains isolated 1962–2021 were used software was developed Java. Results: sequences, including genomes third wave strains, enabled us divide them into three groups: “pre-Haitian”, “Haitian”, “post-Haitian”. All contained toxin-co-regulated tcpACIRS101 pili MARTX rtxA4 cytotoxin with null mutation that caused premature stop codon. However, “pre-Haitian” toxin subunit B classical ctxB1 type prototype histidine kinase always detected, which formed 95 bp long amplicon designated as VCA1095-95. In “Haitian” deletion 8 occurred this gene, shortened 87 (VCA1095-87). Its mandatory combination ctxB7 allele revealed. “post-Haitian” an even shorter rtxA4a due 60 proximal part. Conclusion: Since large revealed strict correlations between certain each group, we consider it possible use only two analysis, i.e. genes. scheme can be facilitate determination newly cultures.
